I could understand the employee’s reaction if the boss had said “Although we can’t give you a salary increase, we’re giving you more responsibilities”. Here, it’s a mystery why the employee would make that sarcastic response.

The only way the sarcastic-sounding answer works for me is if he thinks it’s a trick (or a bad dream, or otherwise not real).
I’m not saying it is funny, but the humor is in the out of place response. You would not expect this response in this type of business interaction from an accomplished business man, even if it actually could logically be used here.
Perhaps he knows that there are already 123 Vice Presidents in his company, that the salary increase is likely to be ten cents an hour or less, and the workload will be doubled. Welcome to corporate America
