Dogs are much more perceptive than people are and always worried that when their person leaves the home that they may not come back. A normal dog would be barking it’s head off at whatever that creature is.
In this case I’d just assume it’s like the TV charaxter “Courage the Cowardly Dog” that notices paranormal things that its owners are oblivious to.

I think the punchline was referring to the wife, and not the dog.
The order I read this comic was interesting. I first read the man and the punchline, then the wife, then the dog. I didn’t notice the hand until after seeing the dog and his expression. Perfect timing.
