I agree with A^2, the cartoonist surely must be referring almost directly to the software by name. (it presents as a website but you must download and install a program to run on your computer)

I mean, I’ve played Second Life, but I don’t expect the general public was super aware of it.
I think this is just a coincidental use of the same phrase. You don’t need to be literally playing Second Life to have a “second life” online.
Second Life was launched in 2003 and had a million users by 2013, so this 2009 comic was published around its period of highest popularity.
I remember hearing about “Second Life,” and got the impression it was like fantasy {name your sport}, but for jobs and relationships rather than sports.
